TLRs constitute of a family of pattern recognition receptors (PRRs) that mediate cellular responses to a large variety of pathogens (viruses, bacteria, and parasites) by specific recognition of so-called ‘pathogen-associated molecular patterns’. Activation of TLRs, a family of at least 11 different members that function either as homo- or heterodimers, leads to activation of NFκB-dependent and IFN-regulatory factor-dependent signaling pathways. TLRs have a central role in innate immunity and are also required for the development of an adaptive immune response. TLRs are expressed by various cells of the immune system, such as macrophages and dendritic cells. TLRs are class I receptors, with a single α-helix that spans the cell membrane. They recognize and respond to molecules derived from bacterial, viral and fungal pathogens, such as lipopolysaccharide (LPS) from the outer membrane of Gram negative bacteria, peptidoglycan fragments from bacterial cell walls and single-stranded and double-stranded RNA from viruses. Toll-like receptor 4 (TLR4; CD284) has been identified, next to MD-2 and CD14, as a receptor that is central to the innate immune response to LPS of Gram-negative bacteria. TLR4 is unique among TLRs in its ability to activate two distinct signaling pathways; one pathway is activated by the adaptors TIRAP (Toll/interleukin-1- receptor (TIR)-domain-containing adaptor protein) and MyD88, which leads to the induction of pro‑inflammatory cytokines. The second pathway is activated by the adaptors TRIF (TIR-domaincontaining adaptor protein inducing interferon‑β) and TRAM (TRIFrelated adaptor molecule), which leads to the induction of type I interferons.

//cdn.shopify.com/s/files/1/0926/3870/8003/files/Hycult_20800x800_20100dpi_fee47646-114e-462d-85d5-583663fd7266.png?v=1753031311
//cdn.shopify.com/s/files/1/0926/3870/8003/files/Hycult_20800x800_20100dpi_53f450c7-5c3e-4b89-836e-d8f776a7abc8.png?v=1753031315
tlr4-human-mab-3c3
TLR4, Human, mAb 3C3

The monoclonal antibody 3C3 reacts with human Toll-like receptor 4 (TLR4, CD284). TLRs belong to a family of proteins that specifically recognize and sense microbial products. They are highly conserved throughout evolution. In Drosophila, toll is required for the anti-fungal response, while the related 18-wheeler is involved in antibacterial defences. TLRs identified as type I transmembrane signalling receptors act as innate immune recognition receptors against many pathogens. TLR4 is a functional receptor for gram-negative bacterial lipopolysaccharides (LPS). TLR4 associates with MD-2 which is absolutely required for LPS-induced activation of TLR4. TLR4 has been identified next to MD-2 and CD14 as a receptor that is central to the innate immune response to LPS of Gram-negative bacteria. The monoclonal antibody 3C3 is a TLR4 function-blocking antibody that is useful for studies on the role of TLR4 as a receptor for LPS induced cytokine production by TLR4 bearing cells.

Applications: Flow Cytometry , Functional Studies

//cdn.shopify.com/s/files/1/0926/3870/8003/files/Hycult_20800x800_20100dpi_88ff853d-8d54-42e4-937c-6c7b4dd27275.png?v=1753031899
//cdn.shopify.com/s/files/1/0926/3870/8003/files/Hycult_20800x800_20100dpi_7d3aa22d-6cd3-4480-a916-ee019bd7e44a.png?v=1753031902
//cdn.shopify.com/s/files/1/0926/3870/8003/files/Hycult_20800x800_20100dpi_ec963f0b-a2cc-4316-9feb-79eb969d8688.png?v=1753031906
//cdn.shopify.com/s/files/1/0926/3870/8003/files/Hycult_20800x800_20100dpi_91e9c336-9133-452e-8be4-794f988dfb92.png?v=1753031909
//cdn.shopify.com/s/files/1/0926/3870/8003/files/Hycult_20800x800_20100dpi_1996213b-6d32-4c10-b84b-874ec4de98e2.png?v=1753031912
//cdn.shopify.com/s/files/1/0926/3870/8003/files/Hycult_20800x800_20100dpi_424cd590-92e0-412e-9c88-6dfa3b817fd1.png?v=1753031916
tlr4-human-mab-4d9
TLR4, Human, mAb 4D9

The monoclonal antibody 4D9 reacts with the Toll-like receptor 4 (TLR4, CD284).
