Exo-spin™ is a modular system that adapts to your changing research needs by providing purification of exosomes from a wide variety of sample inputs for a range of downstream applications. 

Exo-spin™ uses size exclusion chromatography (SEC) columns. A proprietary precipitant buffer may be used to concentrate exosomes before purification on the column. This precipitation is optional for blood, but for other exosome sources, including urine and cell culture media, the samples need concentrating prior to application. Alternative concentrating technologies, such as filtration, may also be used.

Exo-spin™ columns are available with a variety of resin volumes and bed lengths. The shorter bed lengths provide more rapid protocols and lower costs.
For the majority of downstream applications, the 99% purity provided by Exo-spin™ mini, the shortest Exo-spin™ column, provides excellent results.

#Note: Longer bed length improves the resolution, removal of free protein, and purity of exosomes. Purity levels achieved are typical following the protocol provided.

Exosome purification using size exclusion chromatography (SEC)

SEC separates particles in solution based on their size. An SEC column is packed with resin containing stable polymeric spherical beads, each of which has large numbers of nano-scale pores across its surface. The beads leave gaps after packing, the size of which depends on the diameter of the beads. When a solution containing exosomes is added to the top of the resin bed, the smallest particles, including free-proteins, become trapped in the nano-sized pores, slowing their progress, whereas the larger exosomes pass around the outside of the beads and elute first. The very largest particles, beyond the size of the gaps between the beads, are also slowed or stopped completely. The resin used in Exo-spin™ columns has been chosen to specifically allow primary elution of particles in the range 30nm - 250nm. This allows the isolation of exosomes from particles of other sizes.

Exosomes are complex variable structures which require basic characterization as part of any study. Guidance on the minimal level of characterization that should be performed before a study can be published has been provided by the exosome community.  Cell Guidance Systems has developed a range of solutions based on exosome composition and characteristics. These include: exosome marker antibodies, ExoLISA™ detection assay, Instant Exosomes™ and they also offer an NTA size profiling service. 

ExoLISA™ Technology:  Exosome Detection
The ExoLISA™ exosome detection assay is an ELISA-like assay in the sense that it is an antibody sandwich-based assay for the detection of exosome particles. However, there are some significant differences. Unlike a standard ELISA assay, there is no enzymatic reaction. Rather, the exosome target is detected directly with a Europium labelled-antibody, constituting a time-resolved immunofluorescence assay.

The ExoLISA™ exosome assays provide clear and consistent data quickly from purified or unpurified samples in a convenient 96-well format. The kit is available for marker proteins CD9, CD63, and CD81.

Instant Exosomes

Instant Exosomes™ are a range of high-quality and well characterized freeze-dried exosome samples from human cancer cell lines and stem cells. Exosomes are isolated following a combination of precipitation and size exclusion chromatography (SEC). The exosome samples are characterized for overall protein content, using a BCA assay, expression of commonly expressed exosomal markers (CD9, CD63 and CD81) using the ExoLISA™ exosome detection assay and, particle concentration and size distribution by nanoparticle tracking analysis (NTA) using a Zetaview® instrument from Particle Metrix GmbH.

Instant Exosomes™ are isolated and available from the following human cancer cell lines and stem cells:

        • EX301 Instant Exosomes™ from COLO205, a human colon carcinoma cell line
        • EX302 Instant Exosomes™ from LnCAP, a human prostate carcinoma cell line
        • EX303 Instant Exosomes™ from A375, a human malignant melanoma cell line
        • EX304 Instant Exosomes™ from Mesenchymal Stem Cells (MSCs)
